二、干涉显微镜的技术原理 干涉显微镜、相差显微镜和偏振光显微镜都是将光源发射的双光束用标本细节加以 干扰,使之出现光程差。以此对标本细节作某些物理化学和形态学分析。但是这三种光学 成像原理近缘性技术,各有各的特点。 相差显微镜是透过标本细节的衍射光和透过周围介质的直射光之间,对直射光单独 地或更多地加以干扰,从而加大两光束之间的程差。而且相差显微镜只能解决活细胞或未 染细胞的形态学观察。因此相差显微镜的调试和使用比较简便。相差显微镜观察技术中对 于标本制备、玻片等方面要求不十分严密。因此在成像效果方面可能出现细胞边缘的晕昏 现象。 偏振光显微镜是两偏振光束透过标本细节时,由于标本的光学特性而出现折射率的 变化 ,从而引出的光程差观察干涉波的振幅差。因此应用偏振显微镜分析物体的折射率、 光轴方向、厚度等方面远远优于相差显微镜。 干涉显微镜是将两束偏振光中一束通过标本细节,另一束通过周围介质,进而使两光 束满足相干条件,以其光程差的变化分析细胞的组分的净重等更为精确洌定的显微镜。因 此在干涉显微镜技术中,对于标本的大 小、分布密度、厚度要求极严密 。对于所 有光具组、玻片、波晶片的对光轴(显微 镜的光路光轴)顷斜与否要求准确。对载 玻片的质量(均匀性)表面的平滑 ,对盖 玻片的厚度、均质,对于集光镜、物镜的 质量,均有严格要求。干涉显微镜结合显 微分光光度计的光密度扫描技术时,从干涉条纹的移动可以提高测试度,达到士 1 /200波 长(两条干涉条纹间距的实际长度)。由此可以说,利用干涉显微镜的最高分辨本领,用光 的波长(单色光)为标尺,可分辨2nm 的精度 。 图12- 1 1所示,双光束干涉显微镜 y 的光路中透过起偏器的一束线偏振光 在 A 处分解为 ABD 和 ACD 两束线 偏振光。而后在 D 处(前节的 Qz)这两 束光又合成一束偏振光(图 12-11八如 。 果将两束光的光程差调整到扣= 入/2 M 或=亢时,合成波将变成图12-12,1波 和 2波那样相消光束。在光束 ACD .J-. -a 加进折射率甚微的标本 M(图 12-12,4 波)时,ACD 光束的位相(光程)稍许 滞后(图12-12,2波入这时由原来的全暗(1和 2波相消)视野将会变成较弱的亮视野。ACD 8 5 A 、 ... .. .,, . 一 M c 图 12-11 双光束干涉显微镜光路略图 x 田 12- 1 2 双光束干涉光束的正弦曲线 • 126 •
